Project Description:

My main goal is to shed light on lipid dynamics in the plasma membrane of B. subtilis. Lipid dynamics has so far mostly been studied in eukaryotic membranes, where fast spectroscopy techniques like fluorescence correlation spectroscopy (FCS) have revealed that lipid diffusion in membranes is highly heterogeneous. It particularly revealed that certain lipid species transiently assemble and disassemble, a process thought to play a significant role for instance in cellular signalling. In comparison, fewer studies have been performed in prokaryotes and the dynamics of lipids in bacterial membranes remains widely unknown. My project therefore consists in using fluorescence microscopy in general and FCS in particular to measure the diffusion behaviour of the lipids contained in bacterial membranes.
